Job Description:

GT Biosciences is seeking a mission-driven, multidisciplinary Nucleic Acid Scientist to join our Deliveromics Platform Development team. In this role, you will design and optimize novel nucleic acid constructs and chemistries, spanning mRNA, ASO, and siRNA modalities, for targeted delivery. You will support Deliveromics in vivo screening and rigorously evaluate performance in both in vitro assays and animal models. You will independently plan and execute experiments, analyze complex data, and clearly communicate results. As one of the earliest scientific hires at an emerging biotech, you will help shape our platform, collaborate across disciplines, and contribute directly to transformative delivery solutions for genetic medicines. This is a full-time, in-person position based in Cambridge, MA.

Responsibilities:

  • Lead the design and optimization of nucleic acid constructs, mRNA, ASOs, and siRNAs) to maximize potency, stability, and targeted delivery to specific cell types or tissues.

  • Incorporate sequence design strategies, chemical modifications, and structural elements (e.g., modified nucleotides, backbone chemistries, conjugates) to enhance efficacy and reduce immunogenicity or off-target activity across modalities.

  • Optimize multiplexed screening platforms to evaluate how hundreds of nucleic acid-LNP formulations perform in delivering payloads to specific cell types or tissues.

  • Execute studies at the interface of molecular biology, RNA/oligonucleotide chemistry, and formulation science. Prepare DNA templates for in vitro transcription and produce mRNA using enzymatic IVT. Synthesize or procure ASOs and siRNAs; purify and characterize all nucleic acid constructs and incorporate reporter or sequencing-based readouts as appropriate.

  • Collaborate with Deliveromics platform teams to integrate mRNA, ASO, and siRNA payloads into automated in vivo delivery screens. Coordinate experiments in cell culture and animal models to assess delivery efficiency and functional expression or silencing activity.

  • Work closely with LNP chemists, protein engineers, cell biologists, and automation engineers on the Deliveromics platform team to troubleshoot challenges and integrate new methods into the screening pipeline.

  • Provide expertise on mRNA biology to cross-functional projects.

  • Independently design experimental plans, execute assays, and rigorously analyze data. Oversee any supporting scientists or technicians to ensure high-quality data generation.

  • Analyze and interpret results using quantitative and statistical frameworks. Prepare clear summaries and presentations of findings, providing actionable insights to guide design decisions.

  • Maintain complete and well-organized documentation of protocols, experimental records, and data analyses. Develop SOPs and reports to ensure reproducibility and regulatory compliance.

  • Drive continuous improvement of our nucleic acid delivery platform. Contribute to technology transfer and automation efforts as workflows mature toward scale-up and production readiness.

Qualifications:

Required:

  • PhD in a relevant field (e.g., molecular biology, biochemistry, chemical/biomedical engineering) or equivalent experience. Exceptional candidates with substantial relevant experience will also be considered.

  • In-depth knowledge of nucleic acid design principles across mRNA, ASO, and siRNA modalities, including in vitro transcription methods, oligonucleotide chemistry, and nucleic acid purification. Familiarity with mRNA modifications, ASO chemical modifications, and siRNA design rules is required.

  • Hands-on experience producing mRNA via in vitro transcription and/or working with synthetic antisense or siRNA oligonucleotides, including purification and QC of nucleic acid constructs.

  • Skilled in nucleic acid analytics to assess integrity, purity, and potency of mRNA, ASO, and siRNA constructs.

  • Comfort with RNase-free techniques and aseptic handling is essential.

  • Basic coding skills (e.g., Python, R, or MATLAB).

  • Strong knowledge in AI/ML-guided nucleic acid design

  • Practical experience with cell-based or animal assays to evaluate nucleic acid delivery and functional activity (e.g., reporter assays, RT-qPCR for knockdown or expression, sequencing readouts, flow cytometry, or imaging).

  • Strong analytical and problem-solving skills with meticulous attention to detail. Ability to design controlled experiments and maintain high experimental rigor.

  • Excellent written and verbal communication skills. Ability to write clear reports and present findings. A resourceful, adaptable team player who thrives in a fast-paced, cross-functional environment.

  • Ability to work onsite five days per week at our Cambridge, MA laboratory.

Nice to have:

  • Prior experience working with non-human primate models will be a strong plus.

  • Experiences with drug delivery systems (lipid nanoparticles, polymer nanoparticles, AAV, etc.).

  • Hands-on experience troubleshooting liquid-handling platforms or automated lab systems.
